Differentiation of L6 Muscle Cell


Abstract
Cultured tumor cells can be induced to differentiate by drugs such as retinoic acid and cytosine arabinoside. Also, drugs to modiy signal transduction system can induce the differentiation of tumor cells. In signal transduction system,
phospholipase C
(PLC) plays a central role by delivering inositol triphosphate and diacylglycerol. The findings that PLC isozymes are overexpressed in cancer cell in comparison with normal cell suggest a possible role of PLC isozymes in differentiation and
carcinogenesis.
To provide evidence for a functional role of PLC isozyme during the differentiation, we have examined the distributional patterns of PLC isozymes in L6 cell, rat skeletal muscle cell. L6 cell was induced to differentiation by serum deprivation,
protein
kinase C activator (phorbol 12-myristate 13-acetate : PMA) and G protein nhibitor (pertussis toxin : PTx).
@ES The results were as follows :
@EN 1) The proliferation of L6 cell was prominently inhibited by serum deprivation, PMA and PTx at 5th days.
2) Cellular ATP content in control group did not change with time course. In serum deprivation, cellular ATP contents were nearly asme with the control group. The treatment of PMA and PTx made the increment of cellular ATP contents at 5th days.
3) Serum deprivation, PMA and PTx induced the differentiation characters such as cell fusion and muscle bundle formation.
4) PLC-¥â1, PLC-¥â2, PLC-¥â4 and PLC-¥ã2 did not appear in L6 cell.
5) PLC-¥â3 and PLC-¥ä1 were present in L6 cell. Serum deprivation, PMA and PTx did not change the expression of PLC-¥â3 and PLC-¥ä1 during differentiation.
6) PLC-¥ã1 showed differentiation-dependent expression. PLC-¥ã1 expression was decreased by serum deprivation, PMA, and PTx.
From the above results, we suggest that L6 cell is induced bo differentiate by serum deprivation, PMA and PTx and that PLC-¥ã1 PMA and PTx and that PLC-¥ã1 may play an important role in proliferation and differentiation of L6 cell.
